Biological Background: TRIM5 Function and Localization

  • TRIM5, also known as RNF88 or RING finger protein 88, is a member of the tripartite motif (TRIM) protein family characterized by RING, B-box, and coiled-coil domains. It functions as an E3 ubiquitin ligase involved in antiviral innate immunity.
  • Acts as a capsid-specific restriction factor, blocking infection by non-host-adapted retroviruses (including N-MLV, EIAV, SIVmac, FIV, and BIV) early in the life cycle after viral entry but before reverse transcription. Also serves as a pattern recognition receptor that activates innate immune signaling upon binding to the retroviral capsid lattice, triggering its E3 ligase activity, generating Lys-63-linked polyubiquitin chains via the UBE2V1-UBE2N complex, and ultimately leading to NF-kB and MAPK-responsive gene induction.   • Regulates autophagy by releasing BECN1 from its inhibitors BCL2 and TAB2, and acts as a selective autophagy receptor that targets HIV-1 capsid protein p24 for autophagic destruction. Related references: PMID:25127057
  • Localizes to both the cytoplasm and the nucleus, enabling it to intercept incoming viral capsids in the cytoplasm and possibly modulate host gene expression in the nucleus.
  • Contains key post-translational modifications and structural features: acetylation, phosphorylation, and zinc-binding RING and B-box zinc-finger domains that are essential for its E3 ligase activity and antiviral function.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 100–200 of human TRIM5 (NP_149083.2). Due to the polyclonal nature, the antibody may recognize multiple TRIM5 variants and isoforms; confirm specificity in your target species and cell type.
  • For Western blot, a predominant band at ~56 kDa corresponding to TRIM5alpha is expected in human, mouse, and rat lysates. Optimize sample preparation and loading controls to distinguish this from other TRIM members with similar molecular weights.
  • In ELISA, the antibody can be used as a capture or detection reagent. Titrate carefully and validate linearity and sensitivity with recombinant TRIM5 protein or cell lysates expressing the target.
