Biological Background: TRIM5 Function and Localization

  • TRIM5 (Tripartite motif-containing protein 5), aliases RNF88 and TRIM5alpha, is a member of the TRIM protein family characterized by RING finger, B-box, and coiled-coil domains, and functions as a RING-type E3 ubiquitin ligase.
  • It acts as a capsid-specific restriction factor that blocks retroviral infection early after entry but before reverse transcription, targeting viruses such as N-MLV, EIAV, SIVmac, FIV, and BIV. Related references: PMID:17156811
  • Serves as a pattern recognition receptor for retroviral capsid lattices, triggering innate immune signaling via TAK1-dependent activation of NF-kappa-B and MAPK pathways.
