Biological Background: TRIM27 Function and Localization
- TRIM27 (Zinc finger protein RFP, RNF76) is a tripartite motif-containing E3 ubiquitin-protein ligase with RING finger, B-box, and coiled-coil domains, implicated in proliferation, innate immunity, apoptosis, and autophagy. Related references: PMID:22829933, PMID:24144979, PMID:29688809
- Negatively regulates NOD2 signaling by ubiquitinating NOD2 for proteasomal degradation. Together with MAGEL2, mediates Lys-63-linked ubiquitination of WASHC1 to promote endosomal actin assembly and retrograde transport. Related references: PMID:22829933, PMID:23452853
- Modulates apoptosis and antiviral signaling: ubiquitinates USP7 to regulate RIPK1 deubiquitination and TNF-induced apoptosis; cooperates with USP7 or PTPN11 to ubiquitinate and degrade TBK1, suppressing antiviral responses. Related references: PMID:24144979, PMID:26358190, PMID:29688809

Experimental Guidance and Technical Tips
- The immunogen comprises the N-terminal 1-200 amino acids of human TRIM27, encompassing the RING finger domain critical for its E3 ubiquitin ligase activity. This design may support detection of the functional domain.
- For IHC-P, since TRIM27 is expressed in testis within seminiferous tubules, testis tissue sections could be a suitable positive control. Validate staining with appropriate negative controls.
- In ELISA applications, the polyclonal nature of this antibody may provide sensitive detection of native and recombinant TRIM27. Confirm cross-reactivity in mouse and rat lysates if these species are used.
