Biological Background: TRAIL/TNFSF10 Function and Localization

  • TRAIL (TNF-related apoptosis-inducing ligand), also known as Apo-2L, CD253, or TL2, is a member of the TNF superfamily encoded by the TNFSF10 gene.
  • It binds to death receptors TNFRSF10A/TRAILR1 and TNFRSF10B/TRAILR2, and decoy receptors TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and possibly TNFRSF11B/OPG. Related references: PMID:10549288 PMID:26457518
  • Binding to death receptors triggers apoptosis; decoy receptor interaction may modulate its apoptotic activity and serve as a regulatory mechanism.
  • It is expressed as a type II transmembrane protein but can be cleaved to release a soluble ligand; localized to the cell membrane and also secreted.
  • Widely expressed in tissues, with highest levels in spleen, lung, and prostate.
  • The protein undergoes alternative splicing, phosphorylation, and zinc binding, contributing to its regulation.
  • UniProt lists a molecular weight of approximately 32.5 kDa for the full-length protein; additional isoforms or cleaved products may be detected at lower molecular weights, such as 12 kDa.

  • When performing IF/ICC, include suitable permeabilization and blocking steps; co-staining with organelle markers can help confirm localization.
