Biological Background: TLR2 Function and Localization
- TLR2, also known as Toll/interleukin-1 receptor-like protein 4 (TIL4) or CD282, is a member of the Toll-like receptor family that plays a key role in innate immunity.
- It acts as a pattern recognition receptor, forming heterodimers with TLR1 or TLR6 to recognize bacterial lipoproteins and lipopeptides, thereby initiating signaling cascades (PubMed:17889651, PubMed:21078852). Related references: PMID:17889651 PMID:21078852
- Upon ligand binding, TLR2 recruits adaptor proteins MYD88 and TRAF6, leading to activation of NF-kappa-B and production of pro-inflammatory cytokines (PubMed:10426995, PubMed:10426996). Related references: PMID:10426995 PMID:10426996
- TLR2 recognizes a broad range of microbial components including MALP-2, STF, PSM, and OspA-L from bacteria such as Mycoplasma and Borrelia (PubMed:11441107). Related references: PMID:11441107
- It mediates responses to Mycobacterium tuberculosis products like lipoproteins LprA, LprG, LpqH, PstS1, and HSP70, contributing to host-pathogen interactions (PubMed:19362712, PubMed:15809303). Related references: PMID:19362712 PMID:15809303
- Subcellular localization includes the plasma membrane, cytoplasmic vesicle membrane, phagosome membrane, and membrane rafts, with clustering in lipid rafts upon activation (PubMed:16880211). Related references: PMID:16880211
- TLR2 is highly expressed in peripheral blood leukocytes, particularly monocytes, as well as in bone marrow, lymph node, spleen, and lung, consistent with its immune surveillance role.
- The protein features leucine-rich repeats (LRRs) for ligand recognition, a transmembrane domain, and a TIR cytoplasmic domain; it undergoes glycosylation and disulfide bond formation, with a predicted molecular weight of ~90 kDa.
Experimental Guidance and Technical Tips
- For Western blotting, consider the predicted molecular weight of ~90 kDa when analyzing TLR2 expression.
- For IHC-P, antigen retrieval may improve detection; validate in relevant positive controls like spleen or lymph node.
- Polyclonal antibodies may show lot-to-lot variation; include a standardized positive control in each experiment.
- Cross-reactivity with mouse and rat is noted; verify target specificity in the species of interest.
