Biological Background: TIM-3/HAVCR2 Function and Localization
- TIM-3/HAVCR2 (TIMD-3) is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, primarily expressed on T-helper type 1 (Th1) lymphocytes, regulatory T (Treg) cells, dendritic cells, and natural killer (NK) cells.
- The receptor functions as an immune checkpoint, generally inhibiting T-cell responses by attenuating TCR signaling, blocking NF-κB and NFAT activity, and promoting immunological tolerance. Related references: PMID:14556005, PMID:24337741, PMID:26492563
- TIM-3 also plays a role in macrophage activation and can either suppress or enhance immune responses depending on cellular context. Related references: PMID:11823861, PMID:24825777
- As a receptor for galectin-9 (LGALS9), TIM-3 binding leads to suppression of Th1 responses and apoptosis of antigen-specific T cells, though this interaction has been challenged. Related references: PMID:16286920, PMID:24337741, PMID:23555261
- TIM-3 recognizes phosphatidylserine on apoptotic cells, mediating their phagocytosis by dendritic cells and modulating innate immunity.
- The protein contains Ig-like and mucin domains, is palmitoylated, and undergoes phosphorylation and ubiquitination; it locates primarily at the cell membrane and cell junctions.
- Expression of TIM-3 is upregulated on exhausted T cells in chronic viral infections (HCV, HIV) and in tumor microenvironments, making it a key target in cancer immunotherapy.
Experimental Guidance and Technical Tips
- Based on the immunogen design (aa 22–202 of the extracellular domain), this antibody is suitable for detecting both glycosylated and unglycosylated forms of TIM-3 in WB.
- For ELISA, consider pairing this polyclonal antibody with a monoclonal capture antibody to develop a robust sandwich immunoassay.
- Validate antibody performance in the relevant sample system (e.g., immune cells, tissue lysates) and confirm cross-reactivity across human, mouse, and rat as indicated.
