  • Thymidylate synthase (TYMS) is an essential enzyme in the de novo synthesis of thymidylate, a precursor for DNA replication and repair. Alternative splicing gives rise to multiple isoforms.
  • The enzyme catalyzes the reductive methylation of 2'-deoxyuridine 5'-monophosphate (dUMP) to thymidine 5'-monophosphate (dTMP) using 5,10-methylenetetrahydrofolate as a methyl donor.
  • TYMS localizes to the nucleus, cytoplasm, and mitochondria, including the mitochondrial matrix and inner membrane, supporting thymidylate biosynthesis in multiple cellular compartments.
  • As a phosphoprotein, its activity is regulated by phosphorylation. It also undergoes ubiquitylation and forms isopeptide bonds.
  • Mutations in TYMS are associated with dyskeratosis congenita, a rare genetic disorder characterized by bone marrow failure and predisposition to cancer.
