Biological Background: TGFB2 Function and Localization
- TGFB2 encodes the transforming growth factor beta-2 proprotein, which is cleaved into two chains: Latency-associated peptide (LAP) and mature TGF-beta-2. The proprotein is also known as Cetermin and glioblastoma-derived T-cell suppressor factor (G-TSF).

- The LAP chain maintains TGF-beta-2 in a latent state by non-covalently associating with the mature chain, and its activation is regulated by interactions with milieu molecules such as LTBP1 and LRRC32/GARP in the extracellular matrix.
- Following activation, mature TGF-beta-2 signals through binding to TGF-beta receptors (TGFBR1 and TGFBR2), which transduce downstream signals.
- The protein is secreted and localizes to the extracellular space and extracellular matrix.

Experimental Guidance and Technical Tips
- Since the antibody is raised against a recombinant fragment spanning the proprotein region, it may detect both pro-TGFB2 and mature TGF-beta-2, depending on sample preparation and activation state. Consider reducing and non-reducing conditions in WB.
- For ELISA, validate the antibody’s reactivity with the recombinant immunogen and native protein in relevant biological samples (e.g., cell culture supernatants, serum).
- Cross-reactivity with mouse TGFB2 is reported; verify performance in mouse samples if used for cross-species studies.
