Biological Background: TFF1/pS2 Function and Localization
- TFF1 (trefoil factor 1, also known as pS2, BCEI, HPS2, HP1.A, pNR-2) is a small secreted protein of 84 amino acids (9.15 kDa), belonging to the trefoil factor family.
- It functions as a stabilizer of the mucous gel overlying the gastrointestinal mucosa, providing a physical barrier against noxious agents.
- May also inhibit the growth of calcium oxalate crystals in urine.
- Secreted protein found in various mucosal surfaces, with highest expression in the upper gastric mucosal cells; also detected in goblet cells of the intestines, rectum, esophagus, sublingual gland, trachea, pancreatic ducts, gallbladder, vagina, and respiratory system.
- Strongly upregulated in breast cancer tissues and regulated by estrogen in MCF-7 cells, while normal mammary tissue shows low expression.
- In the gastrointestinal tract, highly expressed in normal gastric mucosa and regenerative tissues around ulcers, but expression is often lower in gastric cancer.
- Contains disulfide bonds and is classified as a growth factor; its 3D structure has been determined.

- In IP and IF-P experiments, validate antibody performance with appropriate cellular or tissue models, and optimize fixation and permeabilization conditions for secreted proteins.
