Biological Background: TCEB2 Function and Localization
- TCEB2 encodes Elongin-B (Elongin 18 kDa subunit, SIII p18), a subunit of the transcription elongation factor SIII (elongin) and a core component of multiple cullin-2/5-RING E3 ubiquitin ligase complexes (ECS). The protein is also known as RNA polymerase II transcription factor SIII subunit B and transcription elongation factor B polypeptide 2.
- As part of the SIII/elongin complex, Elongin-B increases RNA polymerase II transcription elongation past template-encoded arresting sites. The transcriptional activity of subunit A is strongly enhanced by binding to the dimeric regulatory subunits B and C (elongin BC complex). Related references: PMID:7638163
- Elongin-B is a central component of ECS (Elongin-Cullin-SOCS box) ubiquitin ligase complexes, where it couples target-recruitment subunits (e.g., VHL, SOCS box proteins) to Cullin/RBX1 catalytic modules, mediating ubiquitination of diverse substrates. Experimental Guidance and Technical Tips
- The immunogen corresponds to human TCEB2 amino acids 1–118; the antibody is expected to recognize full-length Elongin-B. For Western blotting, whole-cell lysates from human or mouse cell lines are suitable. Due to the small molecular weight (13 kDa), consider using 12–15% SDS-PAGE gels and extended transfer times with small-pore membranes (0.2 µm PVDF or nitrocellulose).
- In immunoprecipitation (IP), the polyclonal antibody may efficiently capture Elongin-B along with its interacting partners (e.g., Elongin-C, cullin-2/5, substrate receptors). Validate enrichment under non-denaturing conditions and confirm co-purification by mass spectrometry or Western blot.
