Biological Background: TAF9 Function and Localization

  • TAF9, also known as TAFII31 and TAFII32, is a subunit of the TFIID basal transcription factor complex, which plays a major role in RNA polymerase II (Pol II)-dependent transcription by recognizing promoters and promoting pre-initiation complex assembly. Related references: PMID:33795473
  • The TFIID complex consists of TBP and TBP-associated factors (TAFs), including TAF1 through TAF13. TAF9 is additionally found in the TBP-free TAFII complex (TFTC), the PCAF histone acetylase complex, and the STAGA transcription coactivator-HAT complex.
  • TAF9 and its paralog TAF9B are involved in both transcriptional activation and repression of distinct but overlapping sets of genes, and TAF9 is essential for cell viability. It may also have a role in gene regulation associated with apoptosis. Related references: PMID:15899866
  • Subcellular localization is predominantly nuclear. Post-translational modifications include acetylation and phosphorylation.
  • The protein is classified under keywords such as DNA-binding, nucleus, phosphoprotein, acetylation, and transcription regulation, reflecting its critical role in gene expression control.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 111–210 of human TAF9 (NP_003178.1). This central region is conserved between human and mouse, supporting the observed cross-reactivity.
  • For Western blotting, consider using whole cell lysates or nuclear extracts from human or mouse cell lines. The predicted molecular weight is 29 kDa, but post-translational modifications may cause slight shifts.
  • In ELISA, direct or indirect coating with recombinant TAF9 or peptide antigen may be employed. Titrate the antibody to determine the optimal working concentration for your specific assay conditions.
  • As a polyclonal antibody, lot-to-lot variability may occur; it is advisable to validate the antibody in your sample system and include appropriate controls.
