Biological Background: TAB1 Function and Localization
- TGF-beta-activated kinase 1 and MAP3K7-binding protein 1 (TAB1), also known as MAP3K7IP1, is a critical adapter protein in innate immunity and stress signaling pathways.
- TAB1 mediates JNK and NF-kappa-B activation and proinflammatory cytokine production in response to TLR and cytokine stimulation. Related references: PMID:22307082 PMID:24403530
- It binds the catalytic domain of MAP3K7/TAK1, inducing its autophosphorylation and activation. Related references: PMID:10838074 PMID:25260751 PMID:37832545
- TAB1 also interacts with MAPK14, promoting its autophosphorylation and downstream signaling. Related references: PMID:11847341 PMID:29229647
- Phosphorylation of TAB1 by MAPK14 inhibits TAK1 activity, forming a regulatory feedback loop. Related references: PMID:14592977
- TAB1 recruits MAPK14 to the TAK1 complex to phosphorylate TAB2 and TAB3 regulatory subunits. Related references: PMID:18021073
- The protein is found in the cytoplasm, cytosol, and at the endoplasmic reticulum membrane.
- Ubiquitously expressed, TAB1 undergoes alternative splicing and post-translational modifications, including glycosylation, phosphorylation, and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within amino acids 405-504 of human TAB1; thus, the antibody may recognize epitopes in this C-terminal region. Consider using peptides or truncated proteins to map the exact epitope.
- For Western blot (WB), the predicted molecular weight is 55 kDa (observed band may vary due to modifications). Validate with appropriate positive controls, such as cell lines stimulated with cytokines or TLR agonists to enhance TAB1 signal.
- In immunofluorescence (IF/ICC), TAB1 exhibits cytoplasmic and ER membrane localization. Permeabilization and fixation should be optimized to preserve these structures.
- For ELISA, the antibody can be used as a capture or detection reagent; titrate antibody concentrations using recombinant TAB1 protein or cell lysates.
