Biological Background: T-bet/Tbx21 Function and Localization
- Lineage-defining transcription factor that initiates Th1 lineage development from naive Th precursor cells by activating Th1 genetic programs and repressing the opposing Th2 and Th17 programs. Related references: PMID:10761931
- Activates transcription of signature Th1 genes such as IFNG and CXCR3 in part by recruiting chromatin remodeling complexes (KDM6B, SMARCA4-containing SWI/SNF, H3K4me2 methyltransferase) to establish permissive chromatin states. Related references: PMID:33296702
- Drives Th1 gene expression also via recruitment of Mediator complex and P-TEFb (super elongation complex) to super-enhancers and associated genes in activated Th1 cells. Related references: PMID:27292648
- Represses Th17 lineage commitment by blocking RUNX1-mediated transactivation of RORC, and suppresses Th2 cytokines (IL-4, IL-5, IL-13) through repression of GATA3 and NFATC2.
- Acts as a repressor of type-I IFN response in IFN-gamma abundant microenvironments, protecting Th1 cells from aberrant amplification.
