Biological Background: T-bet/Tbx21 Function and Localization

  • Lineage-defining transcription factor that initiates Th1 lineage development from naive Th precursor cells by activating Th1 genetic programs and repressing the opposing Th2 and Th17 programs. Related references: PMID:10761931
  • Activates transcription of signature Th1 genes such as IFNG and CXCR3 in part by recruiting chromatin remodeling complexes (KDM6B, SMARCA4-containing SWI/SNF, H3K4me2 methyltransferase) to establish permissive chromatin states. Related references: PMID:33296702
  • Drives Th1 gene expression also via recruitment of Mediator complex and P-TEFb (super elongation complex) to super-enhancers and associated genes in activated Th1 cells. Related references: PMID:27292648
  • Represses Th17 lineage commitment by blocking RUNX1-mediated transactivation of RORC, and suppresses Th2 cytokines (IL-4, IL-5, IL-13) through repression of GATA3 and NFATC2.
  • Acts as a repressor of type-I IFN response in IFN-gamma abundant microenvironments, protecting Th1 cells from aberrant amplification.
  • Required for correct development of natural killer (NK) and mucosal-associated invariant T (MAIT) cells. Related references: PMID:33296702
  • Regulates antiviral B-cell responses and promotes antiviral IgG2a isotype switching.
  • Localizes to the nucleus and is expressed specifically in T-cells; subject to phosphorylation and ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 322-535) of human T-bet/Tbx21; consider that epitope recognition may be conformation-sensitive under certain denaturing conditions.
  • For Western blot, validate in your sample system using reducing conditions; the expected molecular weight is ~58 kDa.
  • For IHC-P, antigen retrieval conditions may require optimization; test both heat-induced citrate and EDTA buffers to achieve robust nuclear staining.
  • For intracellular flow cytometry, ensure effective permeabilization and fixation protocols; use appropriate isotype controls to assess non-specific binding.
  • Cross-reactivity is reported for human; reactivity with other species should be confirmed experimentally if needed.
