Biological Background: Syntaxin-4 Function and Localization
- Syntaxin-4 (STX4), also known as renal carcinoma antigen NY-REN-31, is a member of the syntaxin family of SNARE proteins and functions as a plasma membrane t-SNARE.
- It mediates docking of transport vesicles and is essential for the translocation of SLC2A4 (GLUT4) from intracellular vesicles to the plasma membrane in response to insulin.
- In neurons, syntaxin-4 is recruited to neurite tip membrane domains rich in 1-oleoyl-2-palmitoyl-PC (OPPC), facilitating surface expression of the dopamine transporter SLC6A3/DAT.
- Syntaxin-4 is required for normal hearing, as evidenced by its role in docking/fusion events at stereocilia (PubMed:36355422). Related references: PMID:36355422
- Subcellularly, it localizes to the cell membrane, cell projections including neuron projections and stereocilia, and the perinuclear cytoplasm.
- It is expressed in neutrophils and neutrophil-differentiated HL-60 cells, with expression increasing upon differentiation.
