Biological Background: SUFU Function and Localization
- SUFU is a negative regulator of the Hedgehog signaling pathway, sequestering GLI transcription factors in the cytoplasm. Related references: PMID:10559945 PMID:10564661 PMID:10806483
- Upon Hedgehog activation, GLI proteins dissociate from SUFU and translocate to the nucleus to activate target genes. Related references: PMID:24311597 PMID:28965847 PMID:35831023
- In the absence of signaling, SUFU recruits GSK3B to the SUFU-GLI3 complex at cilia, promoting the processing of full-length GLI3 into its repressor form. Related references: PMID:24311597 PMID:28965847
- SUFU may also negatively regulate beta-catenin signaling and is essential for embryonic development, hair follicle formation, and epidermal differentiation.
- The protein localizes predominantly to the cytoplasm, nucleus, and cilia.
- SUFU is ubiquitously expressed in adult tissues, with isoform-specific patterns in fetal development.
- SUFU undergoes post-translational modifications including acetylation and phosphorylation, and is implicated in ciliopathies and tumor suppression.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 325–484 (C-terminal region); consider verifying recognition of relevant SUFU isoforms or processed forms in your model system.
- For Western blot, the predicted molecular weight is 54 kDa; include appropriate positive controls (e.g., HeLa or HEK293 lysates) and validate the antibody in your sample type.
- For IHC-P and IF/ICC, optimize antigen retrieval and fixation conditions; inclusion of isotype controls and tissues with known SUFU expression (e.g., skin, developing limb) is recommended.
- When using ELISA, pair with a suitable detection system and validate the antibody’s reactivity against recombinant SUFU protein or cell lysates.
- Cross-reactivity has been reported for human, mouse, and rat; confirm performance in other species if required.
