Biological Background: Syntaxin-1A Function and Localization
- STX1A encodes syntaxin-1A, a neuron-specific membrane protein also known as HPC-1, which belongs to the syntaxin family of SNARE proteins and is essential for calcium-dependent exocytosis and endocytosis of hormones and neurotransmitters. Related references: PMID:26635000
- Syntaxin-1A forms the core SNARE complex together with SNAP25 on the plasma membrane and VAMP2 on synaptic vesicles; this complex drives synaptic vesicle fusion with the presynaptic membrane, enabling rapid neurotransmitter release.
- Beyond the central nervous system, STX1A participates in regulated exocytosis in other tissues, such as insulin secretion by pancreatic beta cells, GLP-1 release, and acrosomal exocytosis in sperm. Related references: PMID:23091057
- The protein localizes primarily to the cytoplasmic face of the plasma membrane and is enriched at synapses and synaptosomes; it is also found on secretory vesicles and can be secreted in a soluble form.
- Post-translational modifications include phosphorylation and ubiquitination (isopeptide bond formation), which modulate SNARE complex assembly and stability, and the protein contains a coiled-coil domain critical for its function.
- STX1A is highly expressed in embryonic spinal cord, ganglia, adult cerebellum, and cerebral cortex, with additional expression in heart, liver, fat, skeletal muscle, and kidney.
- Alternative splicing generates multiple isoforms, and rare variants have been implicated in Williams-Beuren syndrome, highlighting the protein's clinical relevance in neurological disorders.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide covering amino acids 1–100 of human STX1A, which includes the N-terminal region; this region is relatively conserved between human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blot, a band around 33 kDa is expected under reducing conditions; consider using brain or neuronal cell lysates as positive controls.
- In IHC-P and IF/ICC, antigen retrieval may be required, and formaldehyde fixation should be optimized; strong synaptic staining is anticipated in neural tissues.
- The antibody may be suitable for ELISA; however, the specific dilution factors should be determined empirically for each application and sample type.
- When interpreting results, note that STX1A exists in multiple compartments (plasma membrane, vesicles, secreted), so fractionation or co-localization studies may enrich specific pools.
