Biological Background: STEAP3 Function and Localization
- STEAP3, also known as Dudulin-2, TSAP6, or pHyde, is a six-transmembrane epithelial antigen of prostate 3 and a member of the STEAP family of metalloreductases.
- It functions as a NADPH-dependent ferric-chelate reductase, transferring electrons across the membrane to reduce Fe3+ chelates (PubMed:26205815). Related references: PMID:26205815
- STEAP3 can also reduce Cu2+ to Cu1+, indicating a broader role in metal ion homeostasis.
- It mediates efficient transferrin-dependent iron uptake in erythroid cells.
- Downstream of p53/TP53, STEAP3 participates in apoptosis and cell cycle regulation.
- STEAP3 is indirectly involved in exosome secretion by facilitating the secretion of proteins such as TCTP (PubMed:15319436, PubMed:16651434). Related references: PMID:15319436 PMID:16651434
- Subcellularly, STEAP3 localizes to the endosome membrane.
- Tissue expression includes adult bone marrow, placenta, liver, skeletal muscle, and pancreas; it is down-regulated in hepatocellular carcinoma.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (amino acids 1-180), which may be accessible in both native and denaturing conditions; consider standard blocking and detection protocols for Western blotting.
- Since the antibody recognizes human and mouse STEAP3, validate species-specific reactivity in your experimental model.
- For ELISA applications, inclusion of recombinant STEAP3 protein as a positive control is recommended.
- As STEAP3 is a multi-pass membrane protein, ensure adequate protein extraction and solubilization (e.g., using mild detergents) for optimal detection in WB.
