Biological Background: SOX10 Function and Localization
- SOX10 is a transcription factor essential for the development and maturation of glial cells, including oligodendrocytes (By similarity).
- It specifically activates myelin genes like DUSP15 and MYRF to promote CNS myelination, and cooperates with MYRF in the myelination program (By similarity).
- SOX10 transcriptionally activates MITF, acting synergistically with PAX3 (PubMed:21965087). Related references: PMID:21965087
- The protein shuttles between the cytoplasm, nucleus, and mitochondrial outer membrane.
- It is expressed in fetal brain and adult brain, heart, small intestine, and colon.
- SOX10 is associated with Waardenburg syndrome, Hirschsprung disease, Kallmann syndrome, and deafness; it undergoes alternative splicing and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen targets amino acids 270-360 in the C-terminal region of human SOX10, which may provide specificity to the antibody. Consider confirming reactivity with endogenous protein in appropriate cell or tissue lysates.
- For western blotting (WB), a band around 50 kDa is expected; validate with positive controls expressing SOX10 and consider using nuclear or whole-cell extracts.
- For immunohistochemistry (IHC-P), standard antigen retrieval techniques may be required; verify staining specificity using known SOX10-expressing tissues such as brain or glial tumors.
- For immunofluorescence/immunocytochemistry (IF/ICC), note that SOX10 can localize to both the nucleus and cytoplasm; include appropriate organelle markers and DAPI counterstaining.
- ELISA performance should be assessed with the specific assay format; the recombinant protein used as immunogen may serve as a positive control for coating.
