Biological Background: SOX10 Function and Localization
- Transcription factor SOX-10 is a member of the SRY-related HMG-box (SOX) family of DNA-binding proteins, with a predicted molecular weight of ~50 kDa (UniProt: P56693).
- It plays a central role in developing and mature glia, specifically activating myelin genes such as DUSP15 and MYRF during oligodendrocyte maturation and CNS myelination.
- SOX-10 acts as a transcriptional activator of MITF, working synergistically with PAX3 to regulate melanocyte development. Related references: PMID:21965087
- Subcellularly, it is found in the cytoplasm, nucleus, and mitochondrial outer membrane, reflecting dynamic regulation.
- Expressed in fetal brain and in adult brain, heart, small intestine and colon, with critical functions in neural crest-derived lineages.
- Clinical relevance: mutations in SOX10 are associated with Waardenburg syndrome, Hirschsprung disease, Kallmann syndrome, and deafness.
- Post-translational modifications include phosphorylation, and the protein undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide covering amino acids 350–450 of human SOX10. This C-terminal region may be well-suited for detecting both phosphorylated and unmodified forms; consider phosphatase treatment if band shifts are observed.
- Suitable for western blot (predicted band ~50 kDa), immunocytochemistry/immunofluorescence, and ELISA. For IF/ICC, permeabilization is required for nuclear and mitochondrial detection. Validate staining in relevant cell lines or tissues.
- Cross-reacts with mouse and rat SOX10, but confirm sequence homology in your species of interest before use.
