Biological Background: Somatostatin Receptor 2 (SSTR2) Function and Localization

  • Encodes the somatostatin receptor type 2 (SSTR2, also known as SRIF-1), a G protein-coupled receptor that binds somatostatin-14 and -28.
  • Couples via pertussis toxin-sensitive G proteins to inhibit adenylyl cyclase, and also stimulates phosphotyrosine phosphatase and phospholipase C.
  • Acts as the functionally dominant somatostatin receptor in pancreatic alpha- and beta-cells, mediating the inhibitory effect of somatostatin-14 on hormone secretion.
  • Inhibits cell growth by enhancing MAPK1/2 phosphorylation and upregulating CDKN1B; also negatively regulates insulin receptor signaling via PTPN6.
  • Stimulates neuronal migration and axon outgrowth, participating in brain development; inactivates SSTR3 upon heterodimerization.
  • Subcellularly located at the cell membrane and in the cytoplasm.
  • Predominantly expressed in pancreas, cerebrum, and kidney, with lower levels in jejunum, colon, and liver. In developing nervous system, expressed in cortex and cerebellum.
