Biological Background: SNX4 Function and Localization
- SNX4, or sorting nexin-4, is a member of the sorting nexin family of proteins, which are characterized by the presence of a phox-homology (PX) domain that facilitates phosphoinositide binding and membrane association.
- SNX4 plays a central role in regulating endocytosis and multiple stages of intracellular trafficking (PubMed:12668730, PubMed:17994011, PubMed:32513819). Related references: PMID:12668730 PMID:17994011 PMID:32513819
- It is specifically involved in the recycling of endocytosed transferrin receptor, preventing its lysosomal degradation and thus maintaining iron homeostasis (PubMed:17994011). Related references: PMID:17994011
- SNX4 contributes to autophagosome biogenesis by mediating the trafficking and recycling of the phospholipid scramblase ATG9A, a key protein required for autophagy (PubMed:32513819, PubMed:33468622). Related references: PMID:32513819 PMID:33468622
- Subcellularly, SNX4 localizes predominantly to the cytoplasmic face of early endosome membranes, where it participates in cargo sorting and vesicle formation.
- Post-translational modifications of SNX4 include acetylation and phosphorylation, which may regulate its activity, localization, or protein interactions.
- The SNX4 gene undergoes alternative splicing, giving rise to multiple transcript variants that may encode distinct protein isoforms with potentially different functions.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody corresponds to a recombinant fragment covering amino acids 321–400 of human SNX4, near the C-terminal region, which may allow recognition of native and denatured protein conformations.
- For Western blotting, prepare whole-cell lysates using standard RIPA or SDS sample buffer; a protein band near 52 kDa is expected under reducing conditions. Consider including a positive control such as HeLa or HEK293T cell extracts, which express endogenous SNX4.
- In immunoprecipitation assays, optimize the antibody-to-lysate ratio and use a compatible bead system (e.g., protein A/G agarose) for rabbit IgGs. Validate recovery by Western blot with a different anti-SNX4 antibody.
- For ELISA applications, test a range of coating antigen concentrations and antibody dilutions using recombinant SNX4 protein or peptide. Ensure the epitope is accessible in the coating format.
- The antibody cross-reacts with mouse SNX4; perform initial validation in both human and mouse model systems to confirm species-specific performance.
- Because SNX4 functions in dynamic membrane trafficking pathways, fixation and permeabilization conditions for immunofluorescence may require optimization, though this application has not been specified for this product.
