Biological Background: SNAP25 Function and Localization

  • SNAP25 (Synaptosomal-associated protein 25) is a 25 kDa t-SNARE protein also known as SUP, RIC4, SEC9, and SNAP-25.
  • Functions as a t-SNARE involved in the molecular regulation of neurotransmitter release by mediating vesicle docking and membrane fusion at the synapse.
  • Associates with proteins involved in synaptic vesicle exocytosis and regulates plasma membrane recycling through its interaction with CENPF.
  • Modulates the gating characteristics of the voltage-dependent potassium channel KCNB1 in pancreatic beta cells.
  • Subcellularly located in the cytoplasm (perinuclear region), cell membrane, synapse, synaptosome, and photoreceptor inner segment.
  • Specifically expressed in neurons of the neocortex, hippocampus, piriform cortex, anterior thalamic nuclei, pontine nuclei, and granule cells of the cerebellum.
  • Undergoes post-translational modifications including palmitoylation and phosphorylation; disease variants are associated with epilepsy and intellectual disability (Keywords: alternative splicing, coiled coil, lipoprotein, phosphoprotein).
