Biological Background: SMYD4 Function and Localization
- SMYD4 is a protein-lysine N-methyltransferase that monomethylates PRMT5, thereby modulating its transcriptional activity. Related references: PMID:38438251
- It may also function as a histone methyltransferase (by similarity).
- SMYD4 contains SET and MYND domains, characteristic of this family of methyltransferases.
- It plays a critical role in cardiac development by acting as an epigenetic regulator, negatively regulating HDAC1 in addition to its methyltransferase activity (by similarity).
- Localizes to both the nucleus and cytoplasm.
- Involved in metal-binding and zinc-finger-mediated interactions, consistent with its zinc-finger domains.
Experimental Guidance and Technical Tips
- For western blotting, expect a band around 89–90 kDa. Validate detection in relevant sample systems.
- In immunofluorescence/ICC, anticipate nuclear and cytoplasmic staining patterns. Use appropriate fixation and permeabilization methods.
- For immunoprecipitation, this antibody may enrich full-length SMYD4. Confirm complex formation under native conditions.
- As a polyclonal antibody, batch-to-batch variability should be considered; include positive and negative controls.
