Biological Background: SMPD4 Function and Localization
- SMPD4 (Sphingomyelin phosphodiesterase 4), also known as nSMase-3, SKNY, NET13, is encoded by the SMPD4 gene and produces isoforms via alternative splicing.
- Catalyzes the hydrolysis of sphingomyelin to ceramide and phosphorylcholine, regulating membrane sphingolipid homeostasis (PubMed:16517606, PubMed:25180167). Related references: PMID:16517606, PMID:25180167
- Plays a role in endoplasmic reticulum organization and function, influencing membrane integrity (PubMed:31495489). Related references: PMID:31495489
- May sensitize cells to DNA damage-induced apoptosis (PubMed:18505924). Related references: PMID:18505924
- In skeletal muscle, mediates TNF-stimulated oxidant production (By similarity).
- Localizes to the endoplasmic reticulum membrane, Golgi apparatus membrane, nuclear envelope, cell membrane, and sarcolemma.
- Widely expressed, with highest levels in heart and skeletal muscle; detected in skeletal muscle at the protein level.
- Associated with intellectual disability (disease variant).
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein corresponding to amino acids 600-800 of human SMPD4 (NP_060421.2). For assay optimization, consider performing epitope mapping or using this region for specificity controls.
- SMPD4 is a multi-pass transmembrane protein with a predicted molecular weight of ~98 kDa. Ensure appropriate membrane protein extraction protocols and include positive controls (e.g., heart or skeletal muscle lysates).
- For Western blot, consider using a reducing agent and heating to ensure full denaturation of the transmembrane domains.
- The antibody is validated for WB, IF/ICC, and ELISA with human and mouse samples. Perform cross-reactivity validation in your specific experimental model.
- For IF/ICC, permeabilization may be required to detect ER/Golgi/nuclear envelope localization; suitable fixation methods should be tested.
