Biological Background: Smac/Diablo Function and Localization
- The DIABLO (SMAC) gene encodes a mitochondrial protein that promotes apoptosis by activating caspases in the cytochrome c/Apaf-1/caspase-9 pathway. It opposes inhibitor of apoptosis proteins (IAPs), including BIRC6/BRUCE, caspase-9, and XIAP/BIRC4. Related references: PMID:15200957 PMID:36758104 PMID:36758105
- Smac/Diablo attenuates the stability and apoptosis-inhibiting activity of XIAP/BIRC4 by promoting its ubiquitination and proteasomal degradation, and it disrupts XIAP/caspase-9 interaction to facilitate caspase-3 activation.
- Alternative splicing produces multiple isoforms; isoform 3 is ubiquitously expressed and may be defective in XIAP down-regulation.
- Subcellularly, Smac/Diablo localizes to the mitochondrion and is released into the cytosol upon apoptotic stimuli.
