Biological Background: SLC25A13 Function and Localization

  • SLC25A13 (citrin, ARALAR2) is a mitochondrial carrier protein of the solute carrier family 25, functioning as an electrogenic aspartate/glutamate antiporter essential for the malate-aspartate shuttle and NADH redox balance across the inner mitochondrial membrane. Related references: PMID:11566871, PMID:38937634, PMID:38945283
  • Substrate exchange occurs via a sequential mechanism in which one substrate binds and dissociates before the counter-substrate binds for transport in the opposite direction. Related references: PMID:38937634
  • In addition to aspartate/glutamate antiport, SLC25A13 mediates mitochondrial uptake of L-cysteinesulfinate in exchange for glutamate and a proton, or can exchange L-cysteinesulfinate with aspartate without proton translocation. Related references: PMID:11566871
  • SLC25A13 is localized to the mitochondrial inner membrane and is highly expressed in liver, with lower expression in kidney, pancreas, placenta, heart, and brain.
  • The protein undergoes post-translational modifications including acetylation, phosphorylation, and methylation, and contains calcium-binding domains.
  • Disease variants in SLC25A13 are associated with intrahepatic cholestasis and other metabolic disorders.

Experimental Guidance and Technical Tips

  • The immunogen covers the N‑terminal 1–300 amino acids of human SLC25A13 (isoform NP_001153682.1). Verify isoform expression in the relevant sample system, as this region may differ in alternative transcripts.
  • For Western blot, a band near 74 kDa is expected. Use mitochondrial‑enriched or whole‑cell lysates from liver (high expression) and validate with appropriate controls.
  • IHC‑P applications should include liver sections as a positive control; optimized antigen retrieval and detection conditions may be necessary. Consider antibody titration to reduce background.
  • Polyclonal antibodies offer sensitive detection in ELISA. Cross‑reactivity with the mouse ortholog is reported; if using mouse models, confirm specificity with appropriate knockout or knockdown controls.
  • As with any experimental procedure, consult relevant literature and validate the antibody’s performance in the specific application and sample type.
