Biological Background: SLC22A4 Function and Localization
- SLC22A4 encodes the ergothioneine transporter (ETT), also known as organic cation/carnitine transporter 1 (OCTN1), a sodium-dependent and pH-dependent symporter that mediates uptake of the food-derived antioxidant ergothioneine. - The transporter also mediates bidirectional transport of acetylcholine, contributing to the non-neuronal cholinergic system and exhibiting anti-inflammatory effects in various tissues. Related references: PMID:22206629 PMID:22569296
- SLC22A4 functions as a low-affinity, sodium-dependent L-carnitine transporter at the mitochondrial membrane, helping to maintain intracellular carnitine homeostasis. Related references: PMID:10215651 PMID:15107849 PMID:16729965
- The protein localizes to the apical and basal cell membranes as well as the mitochondrial membrane, and is widely expressed with highest levels in kidney, trachea, ileum, bone marrow, whole blood, and small intestine. It is also found in testis Sertoli cells and specific immune cell subsets (CD68+ macrophages, CD43+ T-cells).
- SLC22A4 is a multi-pass transmembrane glycoprotein with nucleotide-binding properties, and it belongs to the major facilitator superfamily of solute carriers. The gene has been associated with Crohn's disease susceptibility.

- The immunogen corresponds to a conserved region (aa 42–141) of human SLC22A4, supporting cross-reactivity with mouse and rat orthologs. Sequence alignment across species is advisable to confirm epitope conservation.
- For Western blotting, consider using tissue lysates with high expression, such as kidney, ileum, or whole blood. A band at approximately 62 kDa is expected; conditions may require optimization for membrane protein solubilization and reducing agents.
