Biological Background: SLC22A3/OCT3 Function and Localization
-
SLC22A3/OCT3 is a solute carrier family 22 member 3, also known as extraneuronal monoamine transporter (EMT) or organic cation transporter 3 (hOCT3). It belongs to the SLC22 transporter family and functions as an electrogenic voltage-dependent uniporter that transports a wide range of organic cations, including endogenous amines, cationic drugs, and xenobiotics. Related references: PMID:10196521, PMID:10966924, PMID:12538837
-
The transporter mediates the uptake of monoamine neurotransmitters such as dopamine, adrenaline, noradrenaline, histamine, serotonin, and tyramine, playing a key role in the homeostatic regulation of aminergic neurotransmission in the brain. Related references: PMID:10196521, PMID:16581093, PMID:20858707
-
SLC22A3/OCT3 also transports polyamines (spermidine, putrescine, agmatine), guanidine, and choline, contributing to polyamine homeostasis and cation disposition in various tissues. Related references: PMID:10966924, PMID:12538837, PMID:40972734
-
At the subcellular level, the protein localizes to the cell membrane (apical and basolateral domains), mitochondrial membrane, endomembrane system, and nuclear membrane, indicating complex intracellular distribution.
-
Tissue expression is broad, with high levels in liver, kidney, placenta, and various brain regions (cerebral cortex, cerebellum, hippocampus, medulla oblongata). It is also found in intestine, testis, skeletal muscle, heart, and lung, consistent with its systemic role in organic cation handling.
-
Post-translational modifications include glycosylation, and the mature protein has a predicted molecular weight of approximately 61 kDa.
Experimental Guidance and Technical Tips
-
The immunogen is a recombinant protein corresponding to amino acids 289-349 of human SLC22A3/OCT3. This region is intracellular; thus, the antibody is suitable for detecting intracellular epitopes in western blotting. Consider antigen retrieval methods if using immunohistochemistry.
-
For WB, the predicted molecular weight is ~61 kDa. Glycosylation or other modifications may shift the observed band upward; use appropriate gel percentages (e.g., 8–10% SDS-PAGE) and optimize transfer conditions.
-
This antibody cross-reacts with mouse SLC22A3, enabling its use in rodent tissue or cell lysates. Validate with positive (e.g., mouse kidney, brain) and negative (knockout) controls.
-
In ELISA, the antibody can serve as either capture or detection reagent. Perform checkerboard titrations using known amounts of recombinant SLC22A3/OCT3 protein to determine optimal working concentrations.
-
As a polyclonal antibody, it may offer robust signal from multiple epitopes but can also exhibit increased background. Optimize blocking (e.g., 5% non-fat milk or BSA) and washing steps, and include appropriate controls.
