Biological Background: SHMT2 Function and Localization
- SHMT2 (serine hydroxymethyltransferase, mitochondrial) is a pyridoxal phosphate-dependent transferase that catalyzes the reversible conversion of serine and tetrahydrofolate (THF) to glycine and 5,10-methylenetetrahydrofolate (5,10-CH2-THF), a critical one-carbon donor for purine and thymidylate biosynthesis. - SHMT2 localizes predominantly to the mitochondrial matrix and nucleoid, where it associates with mitochondrial DNA and contributes to mitochondrial translation by generating 5,10-CH2-THF for the modification of tRNA wobble uridine bases. Related references: PMID:18063578, PMID:29452640
- By supplying 5,10-CH2-THF, SHMT2 supports the de novo synthesis of thymidylate within mitochondria, preventing uracil accumulation in mitochondrial DNA and maintaining genomic integrity. Related references: PMID:21876188

- For Western blot (WB), a band at approximately 56 kDa is expected in mitochondrial-enriched lysates.
- In immunohistochemistry (IHC) and immunofluorescence (IF/ICC), mitochondrial staining patterns should be observed; co-staining with mitochondrial markers is recommended for validation.
- For immunoprecipitation (IP), consider using native conditions as the antibody targets a folded domain.
