Biological Background: SHMT1 Function and Localization
-
Serine hydroxymethyltransferase, cytosolic (SHMT1), also known as glycine hydroxymethyltransferase, is a pyridoxal phosphate (PLP)-dependent enzyme that catalyzes the reversible conversion of serine and tetrahydrofolate (THF) to glycine and 5,10-methylene THF, serving as a critical component of the folate cycle and facilitating one-carbon biosynthetic reactions essential for methionine, purine, and pyrimidine synthesis. Related references: PMID:24698160, PMID:30035852, PMID:38996576
-
SHMT1 is predominantly localized in the cytoplasm but also shuttles to the nucleus, where it plays a scaffold role essential for assembling the de novo thymidylate synthesis complex by anchoring DHFR and TYMS to the nuclear lamina and DNA replication sites. Related references: PMID:22235121
-
Beyond its canonical role, SHMT1 catalyzes the cleavage of various 3-hydroxy amino acids, such as L-allo-threonine, L-threonine, and 3-phenylserine, forming glycine and the corresponding aldehyde; it also functions in the carnitine biosynthesis pathway as a hydroxytrimethyllysine aldolase (HTMLA). Related references: PMID:38615009
-
SHMT1 possesses RNA-binding capability, forming complexes that selectively regulate SHMT2 mRNA translation and dynamically modulate cytosolic and mitochondrial serine and glycine concentrations, thereby influencing cellular metabolic status. Related references: PMID:38996576
-
The SHMT1 gene undergoes alternative splicing, producing multiple isoforms that may exhibit distinct tissue-specific expression patterns and functional properties.
-
Post-translational modifications, including acetylation and ubiquitination, are known to regulate SHMT1 activity and stability, as indicated by proteomics identification and UniProt keywords.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 285–444 of human SHMT1, a region that may be conserved across species, supporting the observed cross-reactivity with mouse and rat. Validate reactivity in your sample system if using non-validated species.
- For Western blot, the predicted molecular weight is approximately 53 kDa, but confirm the band pattern using appropriate positive controls and consider possible isoforms arising from alternative splicing.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and fixation conditions, as SHMT1 localization (cytoplasmic and nuclear) may require different permeabilization strategies.
- Due to the polyclonal nature of this antibody, minor lot-to-lot variation may occur; include internal references for consistent quantification across experiments.
