Biological Background: SHIP1 (INPP5D) Function and Localization

  • SHIP1 (Phosphatidylinositol 3,4,5-trisphosphate 5-phosphatase 1), also known as INPP5D, SHIP, SHIP-1, hp51CN, SIP-145, and p150Ship, is a 133 kDa protein encoded by the INPP5D gene.
  • It hydrolyzes PtdIns(3,4,5)P3 to PtdIns(3,4)P2, negatively regulating PI3K pathways. Related references: PMID:10764818 PMID:8723348 PMID:8769125
  • It also hydrolyzes PtdIns(4,5)P3, Ins(1,4,5)P3, and Ins(1,3,4,5)P4. Related references: PMID:10764818 PMID:40969890 PMID:8769125
  • SHIP1 negatively regulates B-cell antigen receptor signaling and mediates FCGR2B signaling to terminate activating immune receptor pathways.
  • It controls myeloid cell proliferation, chemotaxis, mast cell degranulation, immune homeostasis, platelet integrin signaling, and JNK signaling in B-cells, as well as osteoclast precursor proliferation and macrophage function; it is required for endotoxin tolerance.
  • SHIP1 is involved in cell-cell junction control, CD32a signaling in neutrophils, and modulation of EGF-induced phospholipase C activity. Related references: PMID:16682172
  • Subcellular localization: cytoplasm, cell membrane, membrane rafts, cytoskeleton.
  • Tissue specificity: predominantly expressed in immune and hematopoietic cells; high in immature CD34+ cells, low in mature CD33+ cells; majority in T-cells, minority in B-cells.
  • Key features: phosphoprotein with SH2 domain and SH3-binding motif; undergoes alternative splicing; magnesium-dependent hydrolase involved in lipid metabolism and immunity.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide within the C-terminal region (aa 900–1000). This region may be present in some but not all splice variants; verify detection of your target isoform.
  • Predicted molecular weight is ~133 kDa. For Western blot, consider using a 6–8% SDS-PAGE gel and optimize transfer for high molecular weight proteins.
  • Reported cross-reactivity with human, mouse, and rat. However, validate the antibody in your species and tissue of interest.
  • For immunofluorescence applications, expect localization to cytoplasm, cell membrane, and possibly cytoskeleton based on reported subcellular distribution.
  • As a polyclonal antibody, lot-to-lot consistency may vary; include appropriate controls and consider validation with a known positive control lysate.
