Biological Background: SESN2 Function and Localization
- Functions as an intracellular leucine sensor that negatively regulates mTORC1 signaling through the GATOR complex. Related references: PMID:18692468 PMID:25263562 PMID:25457612
- Leucine binding to SESN2 disrupts its interaction with GATOR2, thereby activating TORC1 signaling. Related references: PMID:26449471 PMID:26586190 PMID:35114100
- May negatively regulate protein translation in response to endoplasmic reticulum stress, via mTORC1. Related references: PMID:24947615
- May positively regulate transcription by NFE2L2 of genes involved in the response to oxidative stress by facilitating SQSTM1-mediated autophagic degradation of KEAP1. Related references: PMID:23274085
- May prevent the accumulation of reactive oxygen species through N-terminal alkylhydroperoxide reductase activity; originally reported to contribute to oxidative stress resistance by reducing PRDX1, but this could not be confirmed. Related references: PMID:26612684 PMID:15105503 PMID:19113821
- Subcellular localization: Cytoplasm.
- Widely expressed across tissues.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitination (isopeptide bond/Ubl conjugation); also annotated as an oxidoreductase and reference proteome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 211-480 of human SESN2; consider using full-length recombinant protein or lysates from cells expressing SESN2 for validation.
- For Western blotting, expect a band around 54 kDa; appropriate positive and negative controls are recommended to confirm specificity.
- ELISA may be used for quantitative detection; validate in the relevant sample system (e.g., human, mouse, or rat cell lysates) to ensure performance.
- As SESN2 is widely expressed, a variety of cell lines (e.g., HeLa, HEK293) may serve as suitable model systems.
