Biological Background: Serotonin transporter Function and Localization

  • Sodium-dependent serotonin transporter (SERT), encoded by the SLC6A4 gene, belongs to the solute carrier family 6 and is also known as 5HTT or solute carrier family 6 member 4.
  • SERT mediates high-affinity serotonin reuptake by cotransporting one serotonin molecule with one Na⁺ ion in exchange for one K⁺ ion (and possibly a proton) in an electroneutral cycle, terminating serotonergic signaling (PubMed:10407194, PubMed:12869649, PubMed:21730057).   • The protein also exhibits an uncoupled, serotonin-induced channel-like conductance for monovalent cations (mainly Na⁺), which may influence membrane resting potential or excitability.
  • In the central nervous system, SERT is essential for serotonin homeostasis; in the developing cortex, it controls serotonin uptake in glutamatergic neurons for proper spatial organization of cortical circuits.
  • In the mature brain, SERT acts primarily in brainstem raphe neurons to clear synaptic serotonin.

  • For Western blotting, a band around 70 kDa is expected. Membrane protein extraction protocols and gentle lysis conditions may improve detection.
  • In IHC-P, antigen retrieval steps are often required; optimization of fixation and retrieval is advised.
  • For ELISA, peptide competition assays can be used to confirm antibody specificity.
  • Cross-reactivity with mouse and rat SERT has been observed; however, perform independent validation for non-human species.
