Biological Background: SELENOK Function and Localization

  • SELENOK (Selenoprotein K), also designated SELK, HSPC030, or HSPC297, is a member of the selenoprotein family and incorporates selenocysteine in its sequence.
  • It is critical for calcium (Ca²⁺) flux in immune cells, thereby promoting T-cell proliferation and neutrophil migration.
  • SELENOK participates in endoplasmic reticulum-associated degradation (ERAD) of soluble glycosylated proteins. Related references: PMID:22016385
  • Together with the palmitoyltransferase ZDHHC6, SELENOK mediates palmitoylation of inositol 1,4,5-trisphosphate receptor type 1 (ITPR1) in immune cells, modulating ITPR1 stability and function. Related references: PMID:25368151
  • The protein protects cells from endoplasmic reticulum stress-induced apoptosis. Related references: PMID:20692228
  • When overexpressed in cardiomyocytes, SELENOK provides protection against oxidative stress damage. Related references: PMID:16962588
  • SELENOK localizes predominantly to the endoplasmic reticulum membrane and the cell membrane.
  • It is highly expressed in the heart and is involved in foam cell formation through CD36 palmitoylation and macrophage low-density lipoprotein uptake.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide covering the N-terminal region (amino acids 1-94) of human SELENOK; the resulting polyclonal antibody likely recognizes the full-length protein and possibly N-terminal fragments.
  • For Western blotting, a band at approximately 11 kDa is expected; consider using a low-molecular-weight marker and optimizing transfer conditions for small proteins.
  • Since the antibody cross-reacts with mouse SELENOK, it can be utilized in mouse-derived samples; however, validate in the relevant sample system due to potential species-specific differences.
  • ELISA applications may benefit from peptide-based coating; consider determining optimal antibody dilution through checkerboard titration.
