Biological Background: SBDS Function and Localization
- SBDS (Shwachman-Bodian-Diamond syndrome protein) is a ribosomal maturation factor essential for ribosome biogenesis and translation.
- Together with EFL1, SBDS triggers GTP-dependent release of EIF6 from 60S pre-ribosomes in the cytoplasm, enabling 80S ribosome assembly and recycling EIF6 to the nucleus for 60S rRNA processing and nuclear export.
- It is required for normal levels of protein synthesis and has been implicated in cellular stress resistance, DNA damage response, and cell proliferation.
- SBDS is widely expressed across tissues and localizes to the cytoplasm, nucleolus, nucleoplasm, and spindle.
- Post-translational modifications include acetylation; SBDS is a reference proteome member and is associated with the ribosome biogenesis keyword.
- Mutations in SBDS are linked to Shwachman-Diamond syndrome, a congenital bone marrow failure disorder.
- The protein has a known 3D structure and has been identified through direct protein sequencing and proteomics.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (aa 1–250) of human SBDS, which may recognize both full-length and truncated forms of the protein.
- For Western blotting, a specific band around 29 kDa (predicted molecular weight) is expected; consider using positive controls such as HeLa or HEK293 cell lysates and validate in your sample system.
- Due to cross-reactivity with mouse and rat, this antibody can be used in rodent models; however, confirm reactivity and optimal dilution in your experimental setup.
- In ELISA applications, the antibody may serve as a capture or detection reagent; perform pairing and titer optimization as part of assay development.
- Store the antibody as recommended and avoid repeated freeze-thaw cycles to maintain performance.
