Biological Background: SARS-CoV-2 Spike RBD (N501Y) Function and Localization
- The SARS-CoV-2 spike glycoprotein (S) is a trimeric class I fusion protein that mediates viral attachment and entry into host cells via interaction with the angiotensin-converting enzyme 2 (ACE2) receptor. Related references: PMID:32142651, PMID:32155444, PMID:33607086
- It is synthesized as a precursor protein and cleaved into S1 and S2 subunits; the S1 subunit contains the receptor-binding domain (RBD) that engages ACE2, while the S2 subunit mediates membrane fusion.
- Cleavage of the S2 subunit at the S2’ site by host proteases TMPRSS2 (at the cell surface) or cathepsin L (in endosomes) exposes the fusion peptide and triggers irreversible conformational changes required for viral entry. Related references: PMID:32142651, PMID:32703818, PMID:34159616, PMID:34561887
- The spike protein can use alternative entry receptors, including neuropilin-1 (NRP1) and NRP2, integrins, and asialoglycoprotein receptor 1 (ASGR1), potentially influencing tissue tropism. Related references: PMID:33082294, PMID:33082293, PMID:35150743, PMID:34837059
- Structurally, the stalk domain of the S protein contains three hinges that confer considerable orientational freedom to the head domain, a feature important for receptor engagement. Related references: PMID:32817270
- It is heavily glycosylated and contains multiple disulfide bonds; a palmitoylation site is present in the cytoplasmic tail. The spike protein is located on the virion membrane, host ER-Golgi intermediate compartment membrane, and host cell membrane.
- Keywords also include: viral immunoevasion, inhibition of host innate immune response, and superantigen activity, indicating roles beyond cell entry.
Experimental Guidance and Technical Tips
- This antibody was generated against a synthetic peptide spanning amino acids 450–550 of the SARS-CoV-2 Spike RBD (N501Y mutant) and therefore likely recognizes an epitope within this region; confirm specificity against wild-type and other variants if cross-reactivity data are needed.
- For Western blotting (WB), use reducing conditions and consider deglycosylation to improve detection of the ~141 kDa full-length spike protein; validate with recombinant RBD or spike trimer as a positive control.
- In ELISA applications, this monoclonal antibody can serve as a capture or detection reagent; adequate coating concentration and blocking conditions should be optimized in the relevant sample matrix.
- Due to the critical role of the RBD in host cell attachment, this antibody may be useful for neutralization studies or competitive binding assays, but such uses require independent functional validation.
