Biological Background: SARS-CoV-2 ORF3A Protein Function and Localization
- The ORF3a protein is an accessory protein of SARS-CoV-2, also known as Protein 3a, Protein U274, or Protein X1.
- It plays a key role in viral egress by facilitating lysosomal trafficking of virions (PubMed:33157038, PubMed:33422265). Related references: PMID:33157038 PMID:33422265
- ORF3a forms homotetrameric ion channels (viroporins) in endosomal and lysosomal membranes, potentially inducing lysosomal deacidification to promote safe viral release (PubMed:33157038, PubMed:33422265, PubMed:34158638). Related references: PMID:33157038 PMID:33422265 PMID:34158638
- The protein disrupts host autophagy by binding VPS39 on late endosomes, which blocks autophagosome-lysosome fusion and autolysosome formation (PubMed:33422265). Related references: PMID:33422265
- ORF3a induces RETREG1/FAM134B-dependent reticulophagy by interacting with HMGB1, enhancing HMGB1-BECN1 association and causing ER stress and pro-inflammatory responses (PubMed:35239449). Related references: PMID:35239449
- Subcellularly, ORF3a localizes to the virion, host cell membrane, endoplasmic reticulum membrane, secreted forms, cytoplasm, endosomes, and lysosomes.
- Key biological keywords include glycoprotein, host-virus interaction, ion channel, ion transport, activation of host autophagy, and viroporin activity.
- The protein has a calculated molecular weight of ~31.1 kDa (UniProt), consistent with the observed 31 kDa expected for this antibody.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 100-200 of the ORF3a protein; antibody binding epitopes may be concentrated in this region, which should be considered when designing experiments or interpreting results.
- For Western blotting, the expected band for ORF3a is around 31 kDa; validate specificity using lysates from SARS-CoV-2 infected cells or ORF3a-overexpressing systems.
- In ELISA applications, confirm optimal coating conditions and antibody dilution using recombinant ORF3a protein or defined positive controls.
- As a polyclonal antibody, lot-to-lot consistency may vary; it is advisable to qualify each new batch in the intended application prior to large-scale use.
- The antibody is shipped on wet ice; upon receipt, aliquot and store at -20°C (or as recommended by the manufacturer) to maintain stability.
