Biological Background: SARS-CoV-2 NSP6 Function and Localization
- NSP6 is a cleavage product of the replicase polyprotein 1ab (ORF1ab) encoded by SARS-CoV-2, released by the viral main protease.
- Plays a critical role in the formation and maintenance of double membrane vesicles (DMVs), which are viral replication organelles. NSP6 zippers endoplasmic reticulum membranes and connects to lipid droplets, facilitating DMV biogenesis. Related references: PMID:35551511
- Interacts with host TBK1 without affecting its phosphorylation, leading to decreased IRF3 phosphorylation and inhibition of type I interferon production, thereby contributing to innate immune evasion. Related references: PMID:32979938
- Subcellular localization: host endoplasmic reticulum membrane, DMVs, and perinuclear region, consistent with its role in membrane remodeling.
- Predicted to contain multiple transmembrane helices, enabling ER membrane association and membrane zippering functions.
- Involved in viral immunoevasion and suppression of host innate immune responses; identified as a key factor in SARS-CoV-2 replication and pathogenicity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 236-290 of NSP6 (YP_009725302.1), a region conserved among SARS-CoV-2 isolates. Consider using infected cell lysates or transfected cells expressing NSP6 for validation.
- For Western blot, note that the antibody may detect both the full-length replicase polyprotein (~794 kDa) and the cleaved NSP6 product. Validate the observed band sizes in your experimental system, and include appropriate controls to confirm specificity.
- For ELISA, purified recombinant NSP6 protein or viral lysates can be used as coating antigens. Optimize antibody and antigen concentrations for best signal-to-noise ratio.
- Always titrate the antibody for each application and include positive and negative controls to ensure reliable results.
