Biological Background: SAP18 Function and Localization
- SAP18, also known as Sin3-associated polypeptide p18 or 2HOR0202, is a component of the SIN3-repressing complex, enhancing SIN3-HDAC1-mediated transcriptional repression by directing repressive complexes to core histone proteins.
- As an auxiliary component of the exon junction complex (EJC), SAP18 is deposited at splice junctions on mRNAs and participates in mRNA metabolism and nonsense-mediated decay.
- SAP18 is part of the ASAP and PSAP complexes, which bind RNA sequence-independently, regulate intron excision, and inhibit mRNA processing; these complexes promote apoptosis and disassemble upon apoptotic induction.
- It modulates alternative splicing of apoptotic genes, specifically inhibiting formation of the proapoptotic Bcl-X(S) isoform, thereby linking transcription regulation to apoptosis.
- SAP18 localizes to the nucleus, cytoplasm, and nuclear speckles, consistent with its dual roles in transcription and mRNA processing.
- Ubiquitously expressed, SAP18 is involved in core cellular processes including transcription regulation, mRNA splicing, and apoptosis.
- The protein is subject to acetylation and alternative initiation, with a molecular weight of approximately 18 kDa.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 21–172 of human SAP18, a region critical for its functional interactions; consider this when designing blocking or competition experiments.
- For Western blotting, expect a band near 18 kDa; nuclear or whole-cell extracts are recommended. Validate antibody performance in relevant sample systems.
- In IHC-P, optimization of antigen retrieval and primary antibody dilution is advised; staining may be observed in nuclear and cytoplasmic compartments.
- For ELISA, the antibody can be used to detect SAP18 in cell lysates or purified protein; cross-reactivity with mouse and rat SAP18 should be confirmed experimentally.
