Biological Background: SAFB Function and Localization
- SAFB (scaffold attachment factor B1), also designated HAP, HET, SAF-B, or SAFB1, binds to scaffold/matrix attachment region (S/MAR) DNA and serves as a molecular assembly point for a 'transcriptosomal' complex coupling transcription with RNA processing. Related references: PMID:9671816
- SAFB functions as an estrogen receptor corepressor; it binds the HSP27 promoter and reduces its transcription, thus acting as a negative regulator of cell proliferation. Related references: PMID:12660241
- When associated with RBMX, SAFB binds to and stimulates transcription from the SREBF1 promoter (By similarity).
- Subcellular localization is restricted to the nucleus.
- Ubiquitously expressed, with high levels in the central nervous system and low levels in the liver; also found across a wide range of breast cancer cell lines.
- Undergoes multiple post‑translational modifications, including acetylation, methylation, phosphorylation, and ubiquitin‑like conjugation (isopeptide bond), as indicated by UniProt keywords.
- Categorized as a DNA‑binding, RNA‑binding transcription repressor; alternative splicing yields multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen covers the N‑terminal region (aa 1–200) of human SAFB; this region is largely conserved in mouse and rat, consistent with observed cross‑reactivity.
- For Western blot, a band near 103 kDa is expected; tissue or cell lines with high SAFB expression (e.g., CNS‑derived or breast cancer lines) may serve as positive controls.
- In IHC‑P and IF/ICC, nuclear staining patterns should be observed; validate in the relevant sample system and include appropriate controls.
- ELISA suitability suggests the antibody can detect native or denatured SAFB antigen; titration under the specific assay conditions is recommended.
