Biological Background: S100B Function and Localization
- S100B (S-100 protein beta chain, S100 calcium-binding protein B) is a small, abundant calcium- and zinc-binding protein highly expressed in astrocytes, constituting one of the most abundant soluble proteins in brain.
Related references: PMID:20950652 PMID:6487634 - It weakly binds calcium but tightly binds zinc, with distinct binding sites for each ion on each monomer; potassium antagonizes binding of both divalent cations, especially at high-affinity calcium-binding sites.
Related references: PMID:20950652 PMID:6487634 - Acts as a neurotrophic factor promoting astrocytosis and axonal proliferation, and is involved in the innervation of thermogenic adipose tissue by promoting sympathetic innervation.
- Could assist ATAD3A cytoplasmic processing, preventing aggregation and favoring mitochondrial localization.
Related references: PMID:20351179 - May mediate calcium-dependent regulation of physiological processes by interacting with TPR-containing proteins and modulating their activity.
Related references: PMID:22399290 - Localizes to the cytoplasm and nucleus, and is also secreted.
- Although predominantly a brain protein, S100B is also found in a variety of other tissues.
- Undergoes acetylation post-translational modification.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human S100B protein (amino acids 1-92), suggesting the antibody may recognize the native conformation and linear epitopes. Consider validating in applications requiring native structure detection.
- For Western blot (WB), the predicted molecular weight is approximately 11 kDa; a band near this size may be observed in brain or other S100B-expressing tissues.
- This antibody is listed for immunofluorescence (IF-P), immunohistochemistry (IHC-P), and ELISA; optimization of dilution factors and antigen retrieval conditions is recommended based on specific sample types.
- Cross-reactivity with mouse and rat S100B is stated, making it potentially suitable for studies using rodent models; confirm species reactivity in the relevant experimental system.
