Biological Background: RYBP Function and Localization
- RYBP (RING1 and YY1-binding protein), also known as AAP1, DEDAF, YEAF1, and APAP-1, is a Polycomb group (PcG) protein that functions as a core component of the PRC1-like multiprotein complex.
- As part of the PRC1-like complex, RYBP mediates monoubiquitination of histone H2A at 'Lys-119', contributing to chromatin remodeling and transcriptional repression of target genes, including Hox genes. Related references: PMID:25519132
- RYBP inhibits ubiquitination and subsequent degradation of TP53, thereby regulating transcription of TP53 target genes and playing a role in DNA damage response. Related references: PMID:19098711
- It may regulate ubiquitin-mediated proteasomal degradation of other proteins, such as FANK1, to modulate apoptosis. Related references: PMID:14765135, PMID:27060496
- RYBP functions as a transcriptional repressor of E4TF1 and may also bind DNA directly. Related references: PMID:11953439
- In breast cancer, RYBP is down-regulated and acts as a tumor suppressor by down-regulating SRRM3, reducing growth and metastasis. Related references: PMID:27748911
- Subcellularly, RYBP localizes to the nucleoplasm, nucleus, and cytoplasm, and is widely expressed with highest levels in lymphoid tissues and placenta.
- The protein contains zinc finger domains, undergoes phosphorylation, and is subject to ubiquitination-like modifications (isopeptide bond formation).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide covering the C-terminal region (amino acids 100 to C-terminus) of human RYBP; consider using this peptide as a positive control or coating antigen in ELISA.
- For Western Blot (WB), expect a band at ~25 kDa in whole-cell lysates from human, mouse, or rat cells/tissues; validate with appropriate negative and positive controls in your specific sample system.
- Cross-reactivity with mouse and rat has been observed, but performance in other species or applications should be verified experimentally.
- Optimize antibody dilution for your assay conditions; typical starting dilutions for WB are 1:500–1:2000 and for ELISA 1:1000–1:5000, but these should be determined empirically.
