Biological Background: RUNX1 Function and Localization

  • RUNX1, also known as AML1 or CBFA2, is a transcription factor that forms a heterodimeric complex with CBFB to regulate gene expression.
  • It binds to the core consensus DNA sequence 5'-TGTGGT-3' via its runt domain, with CBFB enhancing DNA-binding affinity allosterically.

  • RUNX1 synergizes with ELF4 to transactivate the IL-3 promoter and with ELF2 to transactivate the BLK promoter. Related references: PMID:10207087, PMID:14970218
