Biological Background: RPS3A Function and Localization
- RPS3A (40S ribosomal protein S3a, also known as eS1) is a component of the small ribosomal subunit, which is a large ribonucleoprotein complex responsible for cellular protein synthesis. Related references: PMID:23636399 PMID:34516797
- It forms part of the small subunit (SSU) processome, the first precursor of the small eukaryotic ribosomal subunit, and participates in early rRNA processing and ribosome biogenesis in the nucleolus.
- The protein is encoded by the RPS3A gene (GeneID: 6189) in humans, with aliases including FTE1 and MFTL, and is evolutionarily conserved.
- Subcellularly, RPS3A localizes to the cytoplasm, nucleus, and nucleolus, consistent with its roles in ribosome assembly, protein synthesis, and nuclear processes.
- Key post-translational modifications include acetylation, ADP-ribosylation, and phosphorylation, which may modulate its function and interactions.
- It may also have a role in erythropoiesis via regulation of the transcription factor DDIT3, as suggested by sequence similarity (By similarity).
Experimental Guidance and Technical Tips
- Since the immunogen is a recombinant protein spanning the full-length human RPS3A (1-264), this polyclonal antibody is expected to recognize multiple epitopes, providing robust detection in denaturing and native assays.
- For Western blot, consider using 5-10 µg of total cell lysate from HeLa, HEK293, or other human cell lines; the predicted band size is approximately 30 kDa.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), validate the antibody in tissues or cells with known high ribosomal protein expression (e.g., liver, proliferating cells).
- For immunoprecipitation (IP) and ELISA, optimize the antibody concentration empirically, as polyclonal antibodies may vary between lots.
- Note that cross-reactivity has been observed with mouse and rat species; confirm specificity in your experimental system using appropriate controls.
