  • RNF20, also known as BRE1A or hBRE1, is an E3 ubiquitin-protein ligase that functions as a key component of the RNF20/40 complex, mediating monoubiquitination of histone H2B at lysine 120 (H2BK120ub1).
  • H2BK120ub1 serves as an epigenetic tag for transcriptional activation and is a prerequisite for subsequent histone H3 methylation at lysine 4 and 79 (H3K4me and H3K79me), positioning RNF20 centrally in the histone code and gene regulation.
  • The RNF20/40 complex cooperates with E2 ubiquitin-conjugating enzymes UBE2A or UBE2B to catalyze H2B ubiquitination, and is required for transcriptional activation of Hox genes.
  • RNF20 localizes predominantly to the nucleus and is recruited to specific gene promoters, such as MDM2, via interaction with p53/TP53, where it acts as a transcriptional coactivator.
  • In cancer cells, RNF20 mediates polyubiquitination and proteasome-dependent degradation of isoform 2 of PA2G4.
