Biological Background: RIG-I Function and Localization
- RIG-I (DDX58) is a cytoplasmic innate immune receptor that senses viral nucleic acids and triggers signaling cascades leading to type I interferon and pro-inflammatory cytokine production. Related references: PMID:15208624, PMID:15708988, PMID:16125763
- Upon binding 5'-triphosphorylated dsRNA or short dsRNA, RIG-I forms filaments and recruits RNF135, which facilitates MAVS-dependent activation of TBK1/IKBKE and phosphorylation of IRF3/IRF7. Related references: PMID:28469175, PMID:31006531
- It recognizes a wide range of RNA viruses (e.g., influenza, measles, HCV) and some DNA viruses (e.g., EBV) through RNA polymerase III transcripts. Related references: PMID:21616437, PMID:21884169, PMID:19631370
- Subcellular localization includes cytoplasm, ruffling membranes, cytoskeleton, and tight junctions.
- Tissue specificity: expressed in vascular smooth muscle cells.
- The protein contains DExD/H-box helicase domain, C-terminal regulatory domain, and tandem CARD domains; it is subject to acetylation, phosphorylation, and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 241–455 of human RIG-I (helicase domain region), so the antibody likely recognizes the helicase or C-terminal portion; consider using it for detecting both endogenous and overexpressed RIG-I.
- For Western blot, the predicted molecular weight is ~107 kDa; include appropriate positive controls such as virus-infected or interferon-treated cell lysates.
- Validate the antibody in your specific sample system, as cross-reactivity may vary.
