Biological Background: Rev-Erbα/NR1D1 Function and Localization

  • Rev-Erbα (NR1D1) is a transcriptional repressor and key component of the circadian clock, directly repressing core clock genes including BMAL1, CLOCK, and CRY1.
  • Its repressive activity is heme-dependent; heme binding stimulates interaction with the NCOR1/HDAC3 corepressor complex to enhance transcriptional repression.
  • Regulates diverse metabolic pathways, including lipid and bile acid metabolism, gluconeogenesis, adipogenesis, and macrophage inflammatory responses.
  • Can also act as a transcriptional activator in certain contexts, such as activation of STAR in ovarian granulosa cells and GJA1 in collaboration with SP1.
  • Binds DNA as either a monomer to RevRE elements or a homodimer to RevDR-2 elements, and functions as a competitive repressor of RORA.
  • Widely expressed with high levels in liver, adipose tissue, skeletal muscle, and brain; expression oscillates in a circadian manner.
  • Subcellular localization includes nucleus, cytoplasm, dendrites, and dendritic spines.
