Biological Background: RBX1 Function and Localization
- RBX1 (also known as ROC1, RNF75) is a core component of multiple cullin-RING E3 ubiquitin ligase (CRL) complexes, which target proteins for ubiquitination and proteasomal degradation, regulating cell cycle progression, signal transduction, and transcription.
Related references: PMID:10230407, PMID:10579999, PMID:11961546 - Together with ARIH1, CRL complexes mediate sequential ubiquitination, where ARIH1 adds the first ubiquitin to substrates.
Related references: PMID:27565346 - As part of the CSA complex, RBX1 ubiquitinates RNA polymerase II (POLR2A) at Lys-1268, a key step in transcription-coupled nucleotide excision repair (TC-NER).
Related references: PMID:32355176, PMID:34526721 - RBX1 is a core component of the Cul7-RING(FBXW8) complex, which targets specific proteins for degradation.
Related references: PMID:35982156 - It also forms a Cul9-RING ligase complex that mono-ubiquitinates p53/TP53.
Related references: PMID:38605244 - RBX1 recruits the E2 enzyme CDC34 to the CRL complex and may stimulate its autoubiquitination; it also promotes neddylation of cullins via UBE2M.
- Involved in ubiquitination of KEAP1, ENC1, and KLHL41, and in concert with ATF2 and CUL3, promotes degradation of KAT5, modulating ATM activation.
- The protein localizes to both cytoplasm and nucleus, is widely expressed, and contains a zinc-finger domain essential for its E3 ligase activity; it is subject to acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1-100 of human RBX1 (NP_055063.1). For WB, consider using whole-cell lysates from human, mouse, or rat cell lines; the predicted molecular weight is ~12 kDa. Use appropriate blocking and detection conditions.
- For ELISA, the antibody may be used for detecting RBX1 in serum or purified protein assays; validate with appropriate positive and negative controls.
- Given RBX1’s central role in ubiquitination, consider using proteasome inhibitors (e.g., MG132) to stabilize ubiquitinated substrates in your experimental setup.
- Optimize antibody dilution for each application and sample type; for cross-reactivity with mouse and rat, verify species-specific sequence homology in the immunogen region.
