- RBMX (RNA-binding motif protein, X chromosome), also known as hnRNP G, is a 42 kDa glycoprotein (p43) encoded by the RBMX gene located on the X chromosome.
- It functions as an RNA-binding protein that regulates pre- and post-transcriptional processes, including alternative splicing, transcription, and mRNA processing.
- RBMX/hnRNP G plays a role in tumor suppression by binding to and stimulating transcription from the TXNIP gene promoter, and in lipid metabolism via activation of the SREBF1 promoter when associated with SAFB.
- It is a component of the supraspliceosome complex and regulates alternative splice site selection, either activating or suppressing exon inclusion; for example, it promotes exon 7 inclusion in SMN2 and represses MAPT/Tau exon 10 splicing.
- Subcellular localization is predominantly nuclear, consistent with its role in transcription and splicing regulation.
- Tissue-specifically, RBMX protein is strongly expressed in oral keratinocytes but weakly detected in oral squamous cell carcinomas, suggesting a potential link to tumor suppression.
- Post-translational modifications include acetylation, methylation, phosphorylation, and ubiquitin-like conjugation, which may regulate its activity and interactions.
- It preferentially binds single-stranded 5′-CC[A/C]-rich RNA motifs as a homodimer and also interacts non-specifically with pre-mRNAs.
