Biological Background: RBFOX2 Function and Localization
- RBFOX2, also known as Fox-1 homolog B, RNA binding protein fox-1 homolog 2, RBM9, HRNBP2, and RTA, is a member of the RBFOX family of splicing regulators.
- It functions as an RNA-binding protein that regulates alternative splicing by binding to the 5'-UGCAUGU-3' motif, preventing U2AF2 recruitment to the 3' splice site.
- RBFOX2 regulates alternative splicing of tissue-specific exons and differentially spliced exons during erythropoiesis.
- It acts as a coregulatory factor of estrogen receptor alpha (ER-alpha), modulating transcriptional activity in conjunction with other RNA-binding proteins.
- Together with RBPMS and MBNL1/2, RBFOX2 activates alternative splicing events in vascular smooth muscle cells, as shown in recent studies. Related references: PMID:37548402
- Subcellularly, it is localized to both the nucleus and cytoplasm, consistent with its roles in transcription, splicing, and mRNA processing.
